Digital PCR: Chip Technology with QuantStudio™ 3D Digital PCR System

At the heart of the QuantStudio™ 3D Digital PCR System is a new and high-density PCR chip. Manufactured from a solid silicon substrate, and etched with consistently-sized nanoscale partitions, the chip enables 20,000 PCR reactions. This leads to more precise sample loading and less reaction dropout compared to the droplet-based approaches. And because the chip structure is rigid, the partition reaction volumes remain fixed, meaning you can easily work with samples of varying DNA length and concentration without affecting greatly sample viscosity. This approach eliminates possible additional sample manipulations and extra workflow steps that would be required to normalize viscosities in droplet-based technologies. The use of a chip enables a streamlined workflow with minimal pipetting steps. Once the sample, assay, and master mix have been combined, the reaction mix is loaded onto the chip, and the chip is sealed with a lid. Once closed, the chip will not be opened again, eliminating any possibility for the release of high-copy DNA amplicons into the surrounding lab environment and minimizing exposure to environmental contaminants. Not every partition on the chip will receive a target molecule. As expected, only those partitions with a target will amplify to detectable level. After amplification, the chip is read by the QuantStudio™ 3D Digital PCR Instrument and the amplification status of each partition is individually determined to calculate the absolute quantitative value of the target being measured. Because each chip is sufficient for one sample, there is no need to batch samples if not required. So running only a couple of chips to follow up on a real-time PCR experiment, is no problem. The QuantStudio 3D chip offers 20,000 partitions that accommodate a single sample and two target measurements using both FAM and VIC- labelled assays. Should you feel that your experiment would benefit from greater than 20,000 digital reactions, you can create a larger "virtual chip" through pooling together measurements from multiple chips. Even a million reactions is no problem as you simply configure the software to calculate everything for you. The QuantStudio 3D Digital PCR Chip makes it easy for you to start doing digital PCR, and it will continue to evolve with your demanding applications.
